Position Overview:The Genomic Medicine Unit (GMU) CMC group at
Sanofi is dedicated to the establishment of best in class
manufacturing platforms to support development of life-changing
advanced cell and gene therapy products, including recombinant AAV
vectors, lentiviral vectors and cell therapy products.This
organization encompasses vector and cell line engineering, upstream
and downstream process development, analytics, and clinical
manufacturing support.TheVectorology group is responsible forthe
design, construction and QC of plasmids used for commercial
production of recombinant AAV, as well as providing vectorology
support to other efforts within the unit. We seek a highly
motivated scientist with hands-on experience in molecular cloning,
mammalian tissue culture and knowledge in AAV biology and molecular
virology to join the team.Key Responsibilities:Independently design
and construct viral vector and gene expression plasmids with
minimal supervisionProduce and QC plasmids to meet sequence, purity
and biosafety requirementsPerform mammalian tissue culture
experiments (transfections, infections) to verify plasmid
functionalityDevelop, maintain and demonstrate advanced knowledge
of principles and techniques in molecular cloning, cell culture,
AAV biology and viral vector productionCommunicate effectively with
internal teams and external CROsDeliver plasmids on pipeline
programs in compliance with aggressive timelinesKeep up to date on
industry knowledge and advancements; provide input on optimization
of process or capabilityManage plasmid inventory; provide input and
appropriate reagents to assist different studiesWrite notebooks and
protocols; author reports to support regulatory filings; present
findings/results in multi-functional team meetingsMentor and
support junior staff and build a culture of support and
collaborationMaintain a functional and safe working environment
while maximizing laboratory efficiencyBasic Qualifications:PhD in
virology, molecular biology, microbiology or related biological
sciences, or a Master's degree with a minimum of 4 years of
relevant industry experience or a Bachelor's degree with a minimum
of 6 years of relevant industry experienceStrong expertise in
molecular cloning of vector plasmids (restriction digestion,
ligation and screening), experience with AAV ITR plasmids a
plus.Expertise in basic molecular biology techniques (DNA/RNA
purification, qPCR, SDS-PAGE, westerns, ELISAs, etc)Skilled with
primer design, Sanger sequencing and sequence analysis software
(next generation sequencing software a plus)Good knowledge of
principles and techniques in AAV biology and molecular
virologyHands-on experience with mammalian tissue culture (adherent
and suspension cells) and viral vector productionAbility to design
complex cloning strategy independently with minimal guidance;
ability to interpret results analytically and troubleshoot
technical problemsPreferred Qualifications:Experience with next
generation sequencing (Illumina or PacBio platform) and NGS data
analysis preferredExcellent communication and presentation skills;
excellent organization and attention to detail; critical and
creative thinkerExperience with regulated mammalian gene expression
systemsKnowledge of multiple viral vector expression platforms and
industrial process developmentWorking knowledge of other molecular
cloning methods, such as Gibson assembly, Golden Gate assembly or
